In DNA replication, helicase unwinds the double stranded DNA which forms a replication fork with leading and lagging strand. The leading strand is one which is from 3' to 5'. The enzyme primase generates short strands of RNA which bind to DNA to initiate DNA synthesis by DNA polymerase. However, DNA polymerase can only synthesize DNA in 5' to 3' direction. Hence, the synthesis of DNA is continuous on the leading strand, as it runs from 3' to 5' which is complementary to 5' to 3'. However, the synthesis of DNA on the lagging strand is in the form of short fragments, also known as okazaki fragments.
