    Three isolation techniques used in laboratories are the pour plate method, the spread plate method, and the streak plate method. The pour play method entails placing the inoculum in a petri dish and pouring melted agar into the dish. The spread plate method involves placing the inoculum on a prepared petri dish of agar and spreading it evenly over the agar. The streak plate is the most common method used. In it, a small sample of the inoculum is collected on a sterile loop and spread over the petri dish, leaving a decreasing amount of inoculum along the streak path so that bacteria are more easily isolated into homogenous colonies.

    The three common lab isolation techniques are: quadrant streak, loop dilution and spread plate. Isolation is used for the separation of microbes from a mixed culture into isolated colonies for the purpose of creating a pure culture. The pure culture contains only one type of known microbe. The Streak Plate Method is most commonly used to isolate pure culture.
