Why did the franks wear multiple layers of of clothing when moving to the hiding spot?

They had very limited space to pack items, since they knew they were going to be leaving their house forever and they would never be able to return. You wouldn't want to waste space in your bags carrying clothes when you could just wear multiple layers and transport them that way. It's an efficient way to bring as much stuff with you as you can.
