By Lennie killing Curly's wife, Lennie is in deep trouble and will pay for it with his life. George and Lennie's dream is over because Lennie is in trouble and cannot escape anymore. Without Lennie, George cannot fufill their dream of owning a farm together and Candy who plans to join in with them dream also will not get the ending he wanted.

It means that they will not be able to stay and work for money to buy the house they were dreaming about. In other words it kinda ruins their dream because Lennie kills her so him and George have to run again.
